Water Soluble Peroxyoamid Reagent: Evalulation for Fluorescent Silica Nano-Particles Chemilumineacence

Chemiluminescence methods, both enzymatic and non-enzymatic, are widely used as an analytical tool in various bio-molecular detection applications. Advantages of using chemiluminscence detection are its practical simplicity, relatively low back ground interference, high sensitivity compared to other optical detection methods. Among the non-enzymatic chemiluminescence reactions, peroxyoxalate chemiluminscence (POCL) is one of the most efficient systems. This work demonstrate the development of a method in which synthetic water soluble peroxyoxamide compound (1) was used for POCL reaction in 90% aqueous solution for detection of fluorescent nanoparticles in nanomolar range. The oxamide reagent (1) was synthesized and characterized through melting points, NMR, and Mass spectroscopy. Aqueous solution of oxamide (1) was used to characterize the covalently linked texas red dye with goat anti-mouse IgG antibody and generic goat IgG antibody on mesoporous silica-particles through luminometer and CCD camera. To improve the detection of fluorescent nano-particels, different reaction conditions were studied for increasing stability and reactivity of oxamide (1). Also, fluorosciene dye doped silica-nanoparticles were synthesized and characterized through SEM, TEM and fluorescent microscopic images. For applications in the bio-detection, the function of these nanoparticles was analyzed through chemilumineacense reaction with oxamide (1) and hydrogen peroxide.
