Temperature Uniformity and DNA Amplification Efficiency in Micromachined Glass PCR Chip

We have fabricated glass-based micromachined DNA polymerase chain reaction (PCR) chips with different heater patterns. Two pieces of Pyrex 7740 glass are thermally bonded together to form the PCR reaction channel. The PCR channel was etched to depth of 100_m, the volume of the channel is about 4_l and the chip size is 22mmX20mm .The platinum thin-film micro heater and the temperature sensor have been integrated on back side of the chip. The effects of different heater patterns on the thermal characteristics of the PCR chip have been investigated by experimental measurement using thermal liquid crystal (TLC). From the result, the DNA amplification efficiency is increased with better temperature uniformity. This result indicated that the Micro-PCR can be integrated with Micro capillary electrophoresis (_CE) without losing the detection signals.
