Photodynamic therapy (PDT) is a novel photochemical process for producing localized tissue necrosis, which involves the activation of a photosensitizing drug in the target tissue with light of a specific wavelength matched to an absorption peak of the photosensitiser in the presence of molecular oxygen. However, the photosensitivity is still a major side effect of PDT in clinical treatment. Micelles as photosensitizer carriers are able to provide many advantages, including improvement of drug solubility, reduction in photosensitivity, long circulation time and tumor targeting. In this study, shell crosslinked (SCK) nanoparticle based on poly(ε-caprolactone)-poly(ethylene glycol)-aldehyde was demonstrated to successful delivery of protoporphyrin IX, a photosensitizer, into cells with less cytotoxicity and high PDT efficacy. Therefore, SCK nanoparticle as a novel carrier may have further applications in photosensitizer delivery for photodynamic medicine.
