A DNA/polycation/polyanion system is being designed, which is supposed to be a potentially improved system in comparison to the traditional gene delivery system (DNA/polycation system). The production of such a DNA/polycation/polyanion system is based on polyelectrolyte layer-by-layer self-assembly technology. After investigating several polycations and polyanions, the system produced with polyethylenimine (PEI) and hyaluronic acid (HA) has been achieved. Some physicochemical properties of this system have been characterized. Also, this DNA/PEI/HA system is found to have ~180% increased transfection efficiency on MCF7 cells than DNA/PEI system.
