Realgar As4S4 has been used in traditional Chinese medicine externally or even orally to treat psoriasis,syphilis and malaria.However,its broader healing effect was documented recently.The possibility to synthesize realgar particles as nanosuspensions brings new therapeutic strategies for cancer treatment.In nanoparticles increased surface to bulk ratio is observed which allows a faster drug release into cells and increased bioavailability.The multiple myeloma cell lines OPM1,RPMI-LR5 and U266 were treated with different suspensions of realgar nanoparticles with diameter 100-230 nanometers.Nanosized realgar demonstrates induced time- and concentration-dependent cell cytotoxicity with IC50 within the range of 0.1-0.3 microgram/ml of As.The IC50 values inversely correlate with specific surface area of realgar particles.Apoptotic(Annexin-V+PI-) and late apoptic(Annexin-V+PI+) cells were observed as well as a decrease of mitochondrial potential.Application of realgar nanoparticles might provide a promissing therapeutic approach for multiple myeloma treatment especially in the context of low systemic damage of realgar in comparison to asenic oxide.
