Polymer nanoparticles have been used as carriers to enhance drug delivery. Among them chitosan is the most commonly used carrier, because it promotes long term of incorporated drugs. In the present study,we used DNA vaccine vector named DR-FM2Gelease to formulate a DNA-nanoparticle complex using chitosan.Chitosan-DNA nanoparticles were prepared using a complex coacervation process and the characteristics of the microparticles were investigated.Stability of the nanoparticles was investigated at different pH values by enzymatic degradation and subsequent gel retardation assay. The effect of DNA concentration on release rate from chitosan microparticles was examined using similar intestinal fluid (SIF) and similar gastro fluid (SGF). Cytotoxicity of chitosan microparticles was measured using MTT dye reduction assay in two cell lines. The characteristics of microparticles were evaluated by SEM (scanning electron microscopy), TEM (transmission electron microscopy, and light microscopy.Subsequently, DRFM2G vaccine vector and also the DNA chitosan nanoparticles were used to vaccinate BALB/c mice.
