A High Throughput Multi-stage, Multi-frequency Filter and Separation Device based on Carbon Dielectrophoresis

We present a multi-stage, multi-frequency carbon dielectrophoresis (CarbonDEP) device for high throughput filtering and separation applications in the health care and biotechnology fields. CarbonDEP refers to the use of carbon electrodes to induce DEP. Up to X high aspect ratio carbon electrode arrays are sequentially embedded in a microfluidic channel and excited with different optimized signals in their frequency and magnitude to obtain X dielectrophoresis-active filters, or a super filter with X stages. Particle separation is achieved by sequentially releasing and collecting the particles previously trapped at different arrays. A programmablemulti-channel signal generator has also been engineered. We have demonstrated trapping of viable, non viable yeast and different sized latex beads by positive and negative 3D DEP traps. Simulation confirms experimental results. High throughput particle separation through multi-stage filtering will be demonstrated by the purity of enriched populations obtained when introducing a mix of 1) viable, 2) non viable yeast, 3) 2 and 4) 8 µm diameter latex to the device and polarizing the different arrays with optimized signals. The work presented has an immediate application for the separation and enrichment of cells contained in a peripheral blood sample with a rapid, defined volume, high-throughput system.
