Neuroimaging Distribution of Water Molecules in Brain

We were able to elucidate and build the map of the water molecules distribution in a brain tissue in hippocampus/lateral vetricle area by means of the injected chromophore bis-N-carbazolyl-distyrylbenzene (BND) and FLIM technique. The BND is highly eficient two-photon absorbing chromophore. The fluorescence lifetime of BND diminishes when the amount of water in the surrounding increases.
We observed different fluorescence lifetime of the injected in vivo chromophore depending on water content in different areas: hippocampus (extracellular fluid), lateral ventricle (choroid plexus) (cerebrospinal fluid); and blood vessels whithin the areas. Moreover, the fluorescence lifetime distribution had been drastically changed in case of water deprivation compared to the normal drinking conditions. Statistical analysis of the investigated samples showed that water content decreased at a border of the hippocampus/lateral ventricle area in a deprived case and increased in blood vessels.
The present research involves a detailed investigation of the mechanism of chromophores – water interaction; visualization of water molecules redistribution in brain caused by dehydration; and as results – preliminary investigation of mechanisms of brain self-protection from water deprivation.
