Nano-Mesosphere Drug Carriers for Localized Cancer Chemotherapy

Aggressive conventional chemotherapy of solid cancers of the breast, lung, prostate, and colorectal area is severely limited by systemic toxic complications. New clinically practical concepts are needed for effective local therapeutic modalities. This paper reports recent preclinical studies using direct intratumoral injection of nano-mesoparticulate drug carriers in animal models of lung and breast adenocarcinomas with metastasis. Data for protein (albumin or casein) particles containing >15 wt% mitoxantrone is presented showing greatly prolonged survival (>300%); with possible “cures” when a neoadjuvant treatment is use (with surgical resection of necrotic tumor within 7-10 days of IT treatment). Clinical studies, especially for tissue-conserving neoadjuvant chemotherapy, may be warranted.
