Microfluidics for multiplexing of core needle biopsies

Image guided biopsies for the procurement of diagnostic tissue has proven to be safe, cost effective and useful. However, the amount of tissue obtained is often diminutive and may be exhausted for conventional diagnostic purposes. With the advent of precision medicine, the need for molecular studies has increased the number of tests being requested. An alternative approach needs to be designed that allows a multiplexed analytic approach without compromising their diagnostic utility. To this end, we have designed, built and tested an approach integrating concepts of cytology management, magnetophoresis and microfluidics which enable the multiplexing of diagnostic cells. This approach uses washes of core biopsies to obtain diagnostic cells while keeping the core biopsy intact. The diagnostic cells themselves are enriched and then transported by magnetophoresis for evaluation by traditional brightfield microscopy. Following this they are eluted from the platform and recovered for downstream molecular studies. This approach is in contrast to traditional protocols that sequentially partition tissue for different studies. The advantage of this proposed approach over current methods includes the recovery of high quality nucleic acids and the ability to capture the maximal amount of extractable informative data from these cells.
