Electrospun nanofibers for Sustained Delivery of Cetylpyridinium Chloride for Buccal Topical Application

The aim of this study was to prepare electrospun polymeric nanofiber mat composed of polyvinylpyrrolidone (PVP-Kollidon® 90F), ammonio methacrylate copolymer type B (PMMA-Eudragit® RS100) and cetylpyridinium chloride (CPC), 10:10:1 (wt/wt%) and its characterization for buccal topical application. The morphology of mats with and without the CPC was analyzed using scanning electron microscopy (SEM). The thermal behaviors of the nanofiber mats were characterized using Differential Scanning Calorimeter (DSC) and the Fourier transform infrared spectroscopy (FTI-IR). The amount of CPC remaining in the PVP/PMMA nanofiber mats was determined using UV-vis spectrophotometer. IR spectra of the nanofibers without cetylpyridinium chloride presented similar spectra to the nanofiber with cetylpyridinium chloride and the DSC profiles of CPC showed a single endothermic peak at 85 0C. In vitro permeation study was done in Franz cell diffusion using pig esophageal mucosa as a model for buccal tissue and phosphate-buffered-saline (pH 7.4) as receptor medium. The SEM images revealed that all of the fibers were smooth and the average diameter of fibers was 200–500 nm. These results suggested that the PVP/PMMA nanofibers had an effect of controlled release of cetylpyridinium chloride and can be suitable applied for buccal topical application.
