Nanoparticles based mucoadhesive system (diskettes), an alternative novel transmucosal (buccal) delivery of Fluoxetine hydrochloride (antidepressant drug, highly first pass effect) was developed. In this study, the drug was encapsulated into Sodium / Calcium salts of copolymer of methyl vinyl ether and maleic anhydride (Gantrez MS-955 mucoadhesive polymer) nanoparticles. The drug bearing nanoparticles were prepared by emulsion solvent (internal phase) evaporation method .The drug : polymer ratio (2:3), emusifier concentration (2.5%) and homogenization speed (25000 rpm) were optimized by overlapping of contour plots (overlay plots). The nanoparticles were compacted into small diskettes (5mm, 50mg) for facilitating oral mucosal (buccal) application. The optimized formulation using 32 factorial design indicated it is possible to develop a novel mucoadhesive system for buccal route (site specific) that could be effectively maintain the drug release at modulated rate compared to oral immediate release / oral liquid dosage forms for better antidepressant response with rapid onset ( Lower Tmax) ,avoid first pass effect to the patients and overall improved bioavailability.
