A Simple Method for Fabrication of Microarrays and Microfluidic Device Using PDMS Stamp

We describe a simple micro-fabrication method by using polydimethylsiloxane (PDMS) stamp on a glass substrate. While the microwells used previously facilitated the cell-to-cell formation by PDMS stamping method, not only should it be carefully controlled to avoid nonuniform patterns by deformation, but it is difficult to create thick patterns that allow the cells to be trapped into the microwells. To overcome this technical challenge, a simple and robust fabrication method is demonstrated to study cell-to-cell interaction. Generally, glass-PDMS-glass structures are relatively difficult and expensive to fabricate. However, based on this technology, we also demonstrate a microfluidic device with glass-PDMS-glass configuration which is very useful for biological applications, especially in cell cultures since it allows for the large glass surface area and the surface can be easily modified.
